a) Sequence tags and sequence annotations were used in Human Genome Project.

The genes which are expressed as RNA were identified by Expressed sequence tags.

The total RNA from the cell is sequenced from the cell and RNA is separated into small fragments. This process is done by sequence annotation.

b) YAC is Yeast Artificial Chromosome. It is a cloning vector that helps artificial chromosomes to be cloned in yeast cells.
