When bacteria and rDNA together incubated in CaCl₂ solution, CaCl₂ makes the DNA bind to bacterial cell wall and passing through the pores of cell wall and coming in contact with the cell membrane. Heat shock from low temperature to suddenly subjecting to high temperature changes the permeability of cell membrane and rDNA is passed into the cytoplasm of bacterial cells.
