DNA polymerase cannot initiate DNA replication on its own. DNA replication is a highly regulated and coordinated process in which DNA polymerase plays a central role in extending an existing DNA strand but cannot start a new strand from scratch.

To initiate DNA replication, a separate enzyme called primase is required. Primase synthesizes a short RNA primer complementary to the DNA template strand.
