In rDNA technology, the first step is isolation of DNA. After cell disruption, the cell extract is treated with an appropriate set of enzymes to remove all the other biomacromolecules. As a result, DNA is purified (with minimum contaminants). By adding chilled ethanol, the soluble form of DNA is precipitated out. The precipitated DNA is collected by spooling, by using a covered glass rod.

(1), (2) and (4) Chilled ethanol also precipitates RNA, proteins and cell wall polysaccharides but by the time chilled ethanol is added in this experiment, all those macromolecules are already removed from the sample.
