Regulation at the Plasma Membrane

  • Controls receptor availability: When a ligand binds to a receptor, endocytosis removes that receptor from the cell surface. This limits how strong and how long signaling can occur at the plasma membrane.
  • Adjusts receptor levels over time: Constant stimulation can lead to receptor degradation, which permanently decreases the cell’s sensitivity to that particular signal.

Regulation by Endocytic Trafficking

  • Defines signaling outcomes: After being internalized, receptors are sorted into different compartments. Receptors sent back to the membrane continue signaling, while others are redirected for degradation.
  • Promotes receptor degradation: Receptors delivered to lysosomes are broken down, reducing or completely stopping the signal.
  • Coordinates multiple pathways: The route a receptor takes—such as clathrin-dependent or clathrin-independent endocytosis—determines whether it will be recycled or degraded.

  • Forms internal signaling hubs: Endosomes do more than transport molecules; they serve as active platforms for signaling, allowing new types of signal processing within the cell.
  • Provides spatial and temporal control: By isolating receptors and signaling proteins inside endosomes, cells gain precise control over when and where signaling occurs.
  • Builds signaling complexes: Endosomal membranes host dynamic complexes of signaling molecules, supporting specialized and prolonged intracellular signaling.
