Blue white colonies selection method helps to differentiate recombinants from non recombinants in a single step based on their ability to produce colour in the presence of a chromogenic substrate. In Recombinants, the inserted DNA  fragment inactivates the lac z gene,as a result β-galactosidase enzyme is not produced ,the recombinant colonies appear colorless.In non recombinants ,the lacz gene is functional thus the   β-galactosidase enzyme is expressed giving rise to  blue colour colonies.(colorless substrate converted to blue color)
