Bacteria can be classified into two groups on the basis of the differences in the cell envelopes and the manner in which they respond to the staining procedure developed by Christian Gram. Those cells that take up the gram stain (primary stain) are Gram positive and the others that do not are called Gram negative bacteria.
The peptidoglycan cell wall is the structure that takes up the stain.
In gram positives, the thick peptidoglycan layer retains the primary stain where as in gram negative bacteria, their thin peptidoglycan wall does not retain the primary stain (Gram stain) but retains the secondary stain.
