by bacteria as part of their defence mechanism. In the year 1963, the two enzymes responsible for restricting growth of bacteriophages in Escherichia coli were isolated. One of it is the restriction enzyme that cuts the DNA at specific sites. Since this enzyme identifies the viral DNA and cuts it, thus restricting the growth of bacteriophage within the host cell, the enzyme is called restriction enzyme. The other enzyme is methylase that adds methyl groups to recognition sites of host DNA and masks them from the host restriction.
