This is due to the change of glutamate to valine.  Sickle cell anemia is indeed caused by a point mutation in the gene that codes for the beta-globin subunit of hemoglobin, a protein found in red blood cells. In individuals with sickle cell anemia, a single nucleotide substitution occurs in the DNA sequence of the beta-globin gene. This mutation results in the replacement of glutamic acid (glutamate) with valine at a specific position in the beta-globin protein chain. This change in amino acid sequence causes the hemoglobin molecules to form abnormal, crescent-shaped, or "sickle" structures under certain conditions, such as low oxygen levels or dehydration.
