In mesophyll cells, phosphoenolpyruvate (PEP) is synthesized from pyruvate and ATP by the enzyme pyruvate phosphate dikinase (PPDK). PPDK catalyzes the phosphorylation of pyruvate to form phosphoenolpyruvate (PEP) in the chloroplasts of mesophyll cells.

During glycolysis, PEP is formed from 2-phosphoglycerate by a dehydration reaction catalyzed by the enzyme enolase in the cytosol.
