In a clone of cells, a single-stranded DNA or RNA attached to a radioactive molecule (probe) is allowed to hybridise with its complementary DNA. It is followed by autoradiography detection. Because the probe does not have complementarity with the mutant gene, the clone with the mutated gene will not appear on the photographic film.
