Restriction endonucleases are enzymes capable of making cuts at specific sites (recognition sites) within a DNA molecule and are hence known as molecular scissors. These enzymes recognise and cut the specific DNA sequences in a certain fashion that generates single stranded overhangs, known as ‘sticky ends,’ on the strand.


(2) DNA ligase catalyzes the joining of a nick within a DNA molecule. A nick is a gap/dent in a dsDNA molecule where one phosphodiester bond between adjacent nucleotides of one of the two strands is missing. DNA ligase seals the nick by formation of a new phosphodiester bond.

(3) and (4) Polymerases are enzymes that catalyze the formation of polymeric macro molecules like DNA and RNA. RNA polymerases and DNA polymerases catalyze the formation of RNA and DNA, respectively, based on the template.
