For sequencing of human genome, the total DNA from a cell is isolated and converted into random fragments of relatively smaller sizes. These fragments were inserted into a vector (e.g., BAC or YAC), and introduced into a suitable host cell (e.g., bacteria or yeast). Then, each time the host cell divides, it replicates the DNA sequence along with its own DNA.  The cloning resulted in amplification of each piece of DNA fragment so that it subsequently could be sequenced with ease.
