In order to force bacteria to take up the plasmid, the bacterial cells should first be made ‘competent’ to take up DNA. This is done by incubating the bacterial cells in a specific concentrated calcium salt solution (divalent cation). It renders the bacterial cell wall permeable to the recombinant vectors.
