Restriction enzyme digestions are performed by incubating purified DNA molecules with the restriction enzyme, at the optimal conditions for that specific enzyme. 

PCR stands for Polymerase Chain Reaction and is used for amplification of gene of interest.

The cutting of DNA by restriction endonucleases results in the fragments of DNA. These fragments can be separated by a technique known as gel electrophoresis.

After cutting the source DNA as well as the vector DNA with a specific restriction enzyme, the cut out ‘gene of interest’ from the source DNA and the cut vector with space are joined with help of ligase enzymes. This results in the preparation of recombinant DNA.
